Overview

Zinc finger protein 224 (ZNF224) is a Krüppel-associated box (KRAB) domain-containing transcription factor belonging to the C2H2-type zinc finger protein family. ZNF224 primarily acts as a transcriptional repressor, interacting with corepressors such as KAP1 and PRMT5 to silence gene expression, notably of genes like aldolase A and the mitochondrial citrate carrier. Alternative splicing generates isoforms like ZNF255, which differ in subcellular localization and functions. ZNF224 modulates crucial cellular processes—promoting oncogenic traits and apoptosis resistance in several cancer types by downregulating tumor suppressor genes, including p21 and p53, via direct promoter binding and through miRNA-mediated mechanisms (notably miR-663a upregulation). Depending on cellular context and interacting partners, ZNF224 can function as either an oncoprotein or a tumor suppressor
