Overview

Zinc finger protein 248 (ZNF248) is a Kruppel-type zinc finger-containing transcription factor with a KRAB domain, mainly localized in the nucleus and cytoplasm of many tissues—including highest abundance in hepatocytes and Leydig cells[8]. ZNF248 modulates transcription by binding DNA at RNA polymerase II regulatory regions, and is implicated in the regulation of diverse biological processes including cell cycle progression, mitosis, embryonic organ development, and cell adhesion[1][2][5][9]. Recent studies reveal ZNF248’s overexpression in several cancers, most notably liver hepatocellular carcinoma, where it promotes tumor invasion and proliferation through the PI3K/AKT signaling pathway, reshapes the tumor microenvironment, and influences immune response and drug sensitivity, marking ZNF248 as a novel immunotherapeutic biomarker and a promising therapeutic target in oncology[2].

Mechanism of action

Drugs that target the PI3K/AKT signaling pathway may interact indirectly by modulating ZNF248 effects. Cabozantinib (multi-tyrosine kinase inhibition) may exploit ZNF248-dependent changes in immune infiltration and cell signaling.
