Overview

Zinc finger protein 277 (ZNF277) is a highly conserved C2H2-type zinc finger transcription factor predominantly located in the nucleus, encoded by the ZNF277 gene on human chromosome 7[7][5][2]. It binds DNA in a sequence-specific manner, likely via RNA polymerase II cis-regulatory regions, and is predicted to be involved in regulating cellular senescence and cell proliferation by repressing expression of tumor suppressors such as p21^WAF1^ and the INK4A/ARF locus, possibly through interactions with Polycomb group protein complexes[1][2]. ZNF277 is selectively expressed in intestinal transit-amplifying cells and is overexpressed in colon cancer, playing a role in oncogenic signaling downstream of β-catenin and potentially serving as a marker of proliferative cells in the gut epithelium[1]. High levels of ZNF277 have been linked to increased tumorigenesis in colon cancer models, while its deficiency leads to increased cellular senescence and upregulation of p21^WAF1^[1]. Diseases associated with ZNF277 include colon cancer, Autism Spectrum Disorder, and Specific Language Impairment[2]. No direct therapeutics or safety concerns are documented for this target as of current knowledge.
