Zinc finger protein 280C (ZNF280C)

Target
ZNF280C
Molecular classification
Transcription factor, Zinc finger protein, DNA damage response protein, Chromatin regulator
01

Overview

Zinc finger protein 280C (ZNF280C) is a C2H2-type zinc finger transcription factor involved in epigenetic regulation and DNA damage response. It binds both transcriptionally active and repressive chromatin, often coinciding with CCCTC-binding factor (CTCF) and the cohesin complex. ZNF280C is essential for maintaining trimethylation of histone H3 at lysine 27 (H3K27me3) at specific genomic loci, particularly via recruitment of the epigenetic repressor SMCHD1. It is functionally important in repressing tumor suppressor genes in colorectal cancer and is associated with worse prognosis when overexpressed. Additionally, ZNF280C helps suppress homologous recombination during the DNA damage response, further linking it to genome stability[1][2][3].

Mechanism of action

Not established for drugs, but mechanistic role involves recruiting SMCHD1 to maintain H3K27me3 and antagonism of CTCF/cohesin-mediated chromatin interactions

03

Biological functions

Transcription regulation (both activation and repression)Maintenance of H3K27me3 epigenetic marksChromatin organizationDNA damage response (specifically, inhibition of homologous recombination)
04

Disease associations

Cancer (notably colorectal cancer)Other (possible roles in DNA repair–associated diseases)
05

Safety considerations

None reported for therapeutic targeting; physiological role in genomic stability suggests potential risks if broadly inhibited
06

Biomarkers

Elevated ZNF280C expression is a negative prognostic marker for colorectal cancer
