Overview

Zinc finger protein 281 (ZNF281) is a C2H2-type zinc finger transcription factor that binds to specific DNA sequences, predominantly acting as a gene expression repressor and regulator of pluripotency and differentiation in embryonic stem cells. ZNF281 is rapidly recruited to DNA lesions following genotoxic stress and plays a critical role in DNA double-strand break repair, principally through facilitating the recruitment of key non-homologous end joining (NHEJ) repair factors such as XRCC4. It also regulates transcription of genes involved in DNA damage response and EMT, thereby influencing tumorigenesis, chemoresistance, and metastasis in various cancers. High ZNF281 expression is associated with poorer outcomes in cancer patients and may serve as a prognostic biomarker. No approved drugs currently directly target ZNF281, but its mechanistic role in genome stability and cancer biology makes it a promising research and potential therapeutic target.
