Overview

Zinc finger protein 282 (ZNF282) is a human nuclear transcription factor belonging to the Krüppel-type zinc finger family. It is characterized by its ability to bind specific double-stranded DNA sequences, playing roles in transcriptional regulation as both a repressor and co-activator. ZNF282 was originally identified for binding to the U5 repressive element (U5RE) of the human T cell leukemia virus type I (HTLV-I) long terminal repeat, suppressing viral expression[1][2][5]. More recently, ZNF282 has been shown to act as a co-activator of estrogen receptor α and as an essential co-activator for the transcription factor E2F1, regulating a subset of E2F1 target genes critical for cell cycle progression in various tumors, including breast cancer and esophageal squamous cell carcinoma[1][2]. Overexpression of ZNF282 is associated with poor prognosis in certain cancers (notably ESCC), acts as an independent prognostic biomarker, and its depletion in cancer models leads to reduced proliferation, migration, invasion, and tumorigenicity[1][2]. ZNF282 is also known as HUB1/HTLV-I U5RE-binding protein 1 and functions primarily as a DNA-binding co-regulator, with a role in negative regulation of transcription and selective gene activation[3][4][5].
