Zinc finger protein 292 (ZNF292) is a large, highly conserved transcription factor in the C2H2 zinc finger family, containing 16 zinc finger domains and expressed primarily in the nucleus. It is critical for normal neurodevelopment—mutations in the ZNF292 gene are associated with neurodevelopmental disorders such as intellectual disability, autism spectrum disorder, and ADHD. As a transcription factor, ZNF292 binds DNA, with notable activity at the growth hormone (GH1) gene promoter in cooperation with other factors, and enhances growth hormone gene expression in somatotrophs. It functions as a tumor suppressor, with loss-of-function mutations linked to colorectal, gastric, and other cancers. Emerging evidence indicates ZNF292 and its circular RNA derivative (circZNF292) also play roles in cell stress responses, cataract formation, and cardiovascular pathology. Its only well-established drug-related activity is in gene-level studies; ZNF292 is not currently a recognized therapeutic drug target, nor are there approved drugs that target it directly.
