Overview

Zinc finger protein 326 (ZNF326) is a nuclear protein that functions as a transcription factor and a core component of the DBIRD complex, which interfaces between core messenger ribonucleoprotein (mRNP) particles and RNA polymerase II. Through its DNA-binding C2H2 zinc finger domains and transcriptional activation domain, ZNF326 regulates gene expression by influencing transcript elongation and alternative RNA splicing, particularly in AT-rich genomic regions. ZNF326 plays a significant biological role in neuronal differentiation and is implicated in oncogenesis, particularly in glioma, where it promotes proliferation and invasiveness of cancer cells by activating the Wnt/\u03b2-catenin signaling pathway and upregulating targets such as HDAC7. High expression of ZNF326 correlates with higher tumor grade and poor prognosis in glioma patients[1][4][5]. ZNF326 has also been linked to retinitis pigmentosa, with broader implications in RNA processing and neuronal function[4]. It integrates with other regulatory proteins in the nucleus, contributing to complex regulation of gene expression and cell fate decisions. Currently, there are no direct drugs or known specific inhibitors targeting ZNF326 in clinical use[4][6][8].
