Zinc finger protein 366 (ZNF366; also known as DC-SCRIPT) is a transcription factor containing 11 C2H2-type zinc finger domains, expressed predominantly in dendritic cells, where it contributes to lineage specification and antigen presentation[1][3]. It acts as a transcriptional corepressor, particularly repressing the activity of estrogen and progesterone receptors (nuclear hormone receptors), but can also activate other nuclear receptors such as RAR and PPAR[1][3]. ZNF366 represses ER and PR activity through association with corepressor complexes involving CtBP1 and histone deacetylases[1][3]. In breast cancer, decreased ZNF366 mRNA expression is linked to worse outcomes in ER/PR-positive tumors, making it an independent prognostic marker, but it is not a current drug target[1][3]. No therapeutic drugs or established safety concerns are reported in relation to ZNF366, and there is no evidence to support that it functions as a classical therapeutic target such as a receptor, enzyme, or transporter[1][3][4].
