Overview

Zinc finger protein 431 (ZNF431) is a member of the Krueppel C2H2-type zinc-finger family and functions as a sequence-specific DNA binding transcriptional repressor. It represses transcription of target genes by recruiting histone deacetylases HDAC1 and HDAC2, notably acting as a specific repressor for the patched 1 (PTCH1) receptor during embryonic development. ZNF431 is implicated in the regulation of the sonic hedgehog (SHH) pathway and is required for proper osteoblast differentiation. Mutations in ZNF431 have been associated with non-syndromic facial clefting and may be linked to certain cancers and myelodysplastic syndrome[1][2][3][5].
