Overview

Zinc finger protein 445 (ZNF445) is a nuclear DNA-binding transcription factor of the KRAB and SCAN domain-containing zinc finger family. It binds to double-stranded methylated DNA at imprinting control regions (ICRs) and coordinates the recruitment of repressive complexes such as KAP1, DNA methyltransferases, and histone methyltransferases[3][4][5]. ZNF445 is critical for the maintenance of parent-specific DNA methylation and gene silencing during early embryonic development, protecting genomic imprints from erasure. It is highly intolerant to loss-of-function, and its deficiency is implicated in multilocus imprinting disorders, transient neonatal diabetes, Temple syndrome, and is a likely cause of some cases of infertility and miscarriage[3][5]. ZNF445 expression is most prominent in early embryonic stages, and also occurs in adult tissues including testis and skeletal muscle[1][5].
