Overview

Zinc finger protein 480 (ZNF480) is a KRAB domain-containing transcription factor with 12 C2H2-type zinc finger motifs, belonging to the large KRAB-zinc finger protein family[1][2][4]. It functions predominantly as a positive regulator of transcription, facilitating gene expression via RNA polymerase II and is localized to the nucleus and cytosol[1][2][7]. Highly expressed in embryonic heart and brain as well as select adult tissues, ZNF480 is implicated in developmental pathways, immune microenvironment regulation, and oncogenic processes including glioma and breast cancer progression[2][4][6]. Recent studies identify its elevated expression as an indicator of poor prognosis in lower-grade glioma, strongly associated with changes in immune infiltration and tumor microenvironment, as well as with methylation status serving as a potential regulatory and biomarker mechanism[4][6]. There are no approved drugs targeting ZNF480, though methylating agents can modulate its expression[4].
