Zinc finger protein 506 (ZNF506) is a human Krüppel-associated box zinc finger protein that acts as a sequence-specific transcriptional repressor and epigenetic regulator. It binds to specific retroviral primer binding site (PBS-Pro) DNA motifs, recruiting co-repressors such as KAP1 and promoting heterochromatin formation via H3K9me3, thereby silencing endogenous retrovirus-related elements and contributing to genome stability[2]. In addition to its canonical role as a transcription factor, ZNF506 is a key modulator of the DNA double-strand break repair pathway, localizing to sites of DNA damage via ATM-mediated phosphorylation and interaction with MDC1, where it facilitates the recruitment of phosphatases and downstream repair factors[1]. Loss or mutation of ZNF506 sensitizes cells to DNA-damaging agents and is associated with altered genome integrity. Overexpression or high activity of ZNF506 is implicated in certain cancers, such as acute myeloid leukemia, where it may contribute to disease progression and serve as a potential prognostic biomarker[2].
