Overview

Zinc finger protein 516 (ZNF516) is a nuclear transcription factor of the Krüppel C2H2-type zinc finger family that binds DNA and regulates the transcription of genes involved in several cellular processes, including cell proliferation, differentiation, apoptosis, and thermogenesis[1][2][5][6]. ZNF516 functions as both a transcriptional activator and repressor but is best characterized as a repressor through its association with the CtBP/LSD1/CoREST corepressor complex, especially repressing genes such as EGFR that are critically involved in cell motility and cancer progression[2][3][7]. Loss or reduced expression of ZNF516 is associated with advanced pathological staging and poor survival in breast cancer and is implicated in other disease states including colorectal cancer (where it is subject to copy number loss) and cervical neoplasia (where promoter hypermethylation has been proposed as a biomarker)[2][3][5]. ZNF516 is also involved in developmental processes such as brown adipose tissue differentiation and cellular responses to replication stress[5][6].
