Overview

Zinc finger protein 518B (ZNF518B) is a DNA-binding transcription factor and chromatin regulator containing three zinc finger domains. It participates in chromatin silencing by recruiting and interacting with histone methyltransferase complexes such as G9A (EHMT2) and EZH2, promoting repressive histone modifications (H3K9 and H3K27 methylation) at specific genomic loci. ZNF518B is involved in establishment of heterochromatin at pericentromeric regions, where it interacts with centromere protein B (CENP-B), HP1, and G9A, thus modulating chromosome segregation and stability[1][2][7]. Up-regulation of ZNF518B is observed in colorectal cancer and supports cellular dissemination and metastasis. Genetic variants are linked to gout and diabetes biomarker studies[2].
