Overview

Zinc finger protein 638 (ZNF638) is a nucleoplasmic protein acting predominantly as a transcriptional cofactor and regulator of gene expression. It binds cytidine-rich sequences in double-stranded DNA utilizing multiple domains, including several C2H2-type zinc finger motifs, and is rapidly induced during adipocyte differentiation. ZNF638 interacts with key transcription factors such as C/EBPβ and C/EBPδ to enhance adipogenic transcriptional programs, principally stimulating the expression of peroxisome proliferator‐activated receptor γ (PPARγ) and promoting adipocyte-specific gene expression. In hepatocytes, ZNF638 participates in de novo lipogenesis via interactions with USP7, influencing the expression of major lipogenic enzymes. It is also involved in alternative splicing by interacting with splicing regulators and localizing to nuclear speckles. ZNF638 is associated with several diseases, including some cancers and metabolic syndromes; however, it is not currently a direct therapeutic target.
