Overview

Zinc finger protein 653 (ZNF653) is a human transcriptional repressor and coregulator containing five C2H2-type zinc finger domains. It primarily acts by repressing the transcriptional activity of several nuclear receptors, including NR5A1 (SF-1), PPARG, NR1H3 (LXRα), NR4A2 (NUR77), ESR1 (ERα), and NR3C1 (glucocorticoid receptor). ZNF653 interacts directly with the ligand-binding domains of these receptors, modulating their activity in gene regulation. Although it belongs to the largest family of putative transcription factors in humans, and serves as a general regulatory cofactor for several nuclear receptor pathways, its specific biological and disease implications are not well characterized. It is primarily classified as a poly-zinc finger transcription factor and does not currently have a recognized role as a therapeutic target. If further disease-specific roles or therapeutic relevance are discovered in future research, additional entries may become relevant. Currently, ZNF653 functions mainly as a transcriptional regulator, not as a classic drug target.
