Zinc finger protein 667 (ZNF667) is a member of the KRAB/C2H2-type zinc finger protein superfamily, functioning as a DNA-binding transcription factor. It possesses a KRAB (Krüppel-associated box) domain at the N-terminus and 14 C2H2-type zinc finger motifs at the C-terminus[1]. ZNF667 is predominantly expressed in the brain and heart, where it localizes to the nucleus and acts mainly as a transcriptional repressor[1][2]. The protein directly binds specific DNA consensus motifs (core sequence 5′-CTTA-3′) to regulate the transcription of downstream genes. Functionally, ZNF667 is upregulated in response to oxidative stress, such as that induced during myocardial ischemia or in ischemic brain injuries; in these contexts, it exerts a cytoprotective, anti-apoptotic effect—mediated, in part, by direct suppression of the pro-apoptotic gene Bax[1][3]. While ZNF667 is not currently recognized as a direct therapeutic target, its regulatory roles in stress response and cell survival pathways may render it of future interest in cardiovascular and neuroprotective therapeutic research[1][3][5][7].
