Overview

Zinc finger protein 687 (ZNF687) is a member of the C2H2-type zinc finger transcription factor family, characterized by its DNA-binding zinc finger motifs and presumed regulatory roles in gene expression[2][5]. ZNF687 is implicated in bone biology, particularly in bone differentiation and development, and is known to play a crucial role in osteoclastogenesis and osteoblastogenesis[2][4][1]. Mutations in the ZNF687 gene are strongly associated with severe forms of Paget disease of bone (especially type 6), often resulting in abnormal bone remodeling and occasionally neoplastic transformations such as giant cell tumor of bone[2][4][1]. Beyond skeletal disorders, ZNF687 has oncogenic roles: its overexpression promotes tumor cell proliferation, invasion, and angiogenesis in lung adenocarcinoma (via the PI3K/AKT pathway) and colorectal cancer (via the Wnt/β-catenin pathway)[2]. As with many zinc finger proteins, ZNF687 contains intrinsically disordered regions that enable liquid-liquid phase separation, potentially compartmentalizing gene expression and signaling events without physical barriers[1]. There is emerging evidence for its use as a biomarker for bone diseases and certain cancers, but no specific drugs or targeted therapies are currently reported for this molecule[2][4][5].
