Overview

Zinc finger protein 750 (ZNF750) is a 723-amino acid nuclear transcription factor characterized by a single degenerate zinc finger domain, which uniquely utilizes a CCHC zinc-binding motif and demonstrates high thermal stability[1][3]. ZNF750 mediates terminal differentiation of keratinocytes by activating skin-specific genes (e.g., in concert with KLF4) and repressing progenitor genes (with proteins such as KDM1A, RCOR1, CTBP1/2)[1][2][5]. Its expression is specific to keratinocytes and is regulated by TP63[5]. Mutations or reduced expression of ZNF750 are implicated in skin pathologies (such as familial psoriasis) and various cancers, where it acts as a tumor suppressor by inhibiting cancer stem cells and controlling cell proliferation, migration, and adhesion[1][2][4][5]. ZNF750 may interact with DNA indirectly, possibly through protein complexes, as its atypical zinc finger is more likely to mediate protein–protein interactions rather than direct DNA binding[1][3]. Experimental upregulation (e.g., via retinoic acid) induces differentiation and reduces cell viability in tumor models, highlighting its potential as a therapeutic target[4].
