Zinc finger protein 777 (ZNF777)

Target
ZNF777
Molecular classification
Transcription factor, KRAB (Krüppel-associated box) zinc finger protein family, DNA-binding protein
01

Overview

Zinc finger protein 777 (ZNF777) is a human transcription factor belonging to the KRAB-ZFP (Krüppel-associated box-containing zinc finger protein) family. It contains a KRAB domain that interacts with transcriptional corepressors and multiple zinc finger domains that bind DNA in a sequence-specific manner, regulating transcription via RNA polymerase II. ZNF777 localizes to the nucleus and is involved in the negative regulation of cell proliferation, particularly in cancer cell biology, where its overexpression can inhibit proliferation by down-regulating FAM129A and inducing cell cycle inhibitor p21. The gene is located on chromosome 7 and expressed in several tissues. Despite its biological relevance, ZNF777 is not currently recognized as a direct target for therapeutic drugs or intervention.

Mechanism of action

Not applicable (no drugs known to target ZNF777).

03

Biological functions

Regulation of gene expression (DNA-templated transcription)Negative regulation of cell population proliferationProtein heterooligomerizationSequence-specific DNA bindingZinc ion binding
04

Disease associations

Cancer (implicated in proliferation of neoplasm cells)Other (potential involvement in cell cycle regulation via interaction with p21 and FAM129A, no direct disease association reliably established in broader literature)
05

Safety considerations

None documented, as ZNF777 is not a direct therapeutic target. Overexpression affects cell proliferation, which could have implications for cancer biology, but no drug-related safety issues are described
06

Biomarkers

May serve as a tumor biomarker due to influence on cell proliferation and interaction with cell cycle proteins, but not widely validatedExpression level changes may reflect cell proliferation state or tumor biology
