Overview

Zinc finger protein 808 (ZNF808) is a primate-specific, KRAB domain-containing zinc finger transcription factor that acts as a transcriptional repressor, primarily targeting the long terminal repeats (LTRs) of endogenous retroviruses, particularly MER11 elements. It regulates gene expression during early pancreatic development by silencing specific transposable elements and preventing inappropriate activation of liver-associated gene expression programs in pancreatic precursor cells. Loss-of-function variants in ZNF808 are associated with non-syndromic neonatal diabetes and pancreatic agenesis in humans, highlighting its critical role in maintaining normal pancreatic endocrine differentiation and development. This gene is predicted to be active in the nucleus and may play additional roles in cell fate specification. Notes regarding therapeutic target status: ZNF808 is not currently a recognized therapeutic target (such as a receptor, enzyme, or transporter amenable to pharmacological modulation) and there are no known drugs interacting with it or mechanisms of action for therapeutic agents targeting this protein. Its primary biomedical relevance is related to loss-of-function genetic disorders rather than therapeutic intervention.
