Overview

Zinc finger protein 831 (ZNF831) is a large, nuclear, DNA-binding transcription factor belonging to the zinc finger C2H2-type protein family. ZNF831 regulates gene transcription, predominantly by binding specific DNA motifs and repressing gene expression. One characterized action is transcriptional repression of STAT3, a key oncogenic driver, leading to decreased expression of downstream anti-apoptotic proteins such as Bcl2, thereby promoting apoptosis in breast cancer. ZNF831 also binds various metal ions (including zinc, iron, calcium, magnesium), a feature typical of zinc finger proteins, and this metal-binding role may link ZNF831 to hematopoietic processes and diseases such as anemia. Reduced expression of ZNF831—often due to promoter hypermethylation—has been observed in several cancers, including breast cancer, and is associated with decreased apoptosis and tumor growth. ZNF831’s activity has also been linked to immune regulation and disease phenotypes such as preeclampsia and inflammatory conditions. Emerging evidence proposes ZNF831 as a promising biomarker and a potential tumor suppressor offering new avenues for targeted therapy[1][2][4][5][7][8].
