Zinc finger protein 93 (ZNF93) is a primate-specific Krüppel-associated box (KRAB) zinc finger protein that acts as a transcription factor and functions primarily as a repressor of transposable elements, notably L1 (LINE-1) retrotransposons. It directly binds to L1 elements to repress their activity, thereby contributing to the maintenance of genome stability. In addition to transposable element silencing, ZNF93 represses the expression of APOBEC3B, a mutagenic enzyme involved in tumorigenesis and DNA replication stress, through both TRIM28-dependent and independent mechanisms. ZNF93 is expressed in proliferating cells, highly correlated with cell proliferation rates, and tends to be overexpressed in various cancer types, where it helps cancer cells adapt and survive by fine-tuning the balance between genomic damage and cell proliferation. Its depletion results in increased replication stress, DNA damage response activation, and decreased proliferation. No known drugs are currently documented to interact directly with ZNF93, nor are any mechanisms of drug action or clinical biomarkers established for this target.
