Zinc finger protein ZIC 2 (ZIC2) mRNA encodes a transcription factor belonging to the ZIC family, characterized by C2H2-type zinc finger domains. It plays a fundamental role during embryogenesis, specifically in the development of the central nervous system, neural tube closure, and the establishment of the left-right body axis (UniProt: Q9UBW0). In clinical pathology, mutations in the ZIC2 gene are a known cause of holoprosencephaly type 5, a severe brain malformation (NIH: GeneReviews). Beyond its developmental role, ZIC2 mRNA is frequently overexpressed in various cancers, including hepatocellular carcinoma and pancreatic cancer, where it functions as an oncogene by driving cell cycle progression and inhibiting apoptosis (PubMed: 28656245). Therapeutic strategies targeting ZIC2 mRNA typically involve the use of small interfering RNAs (siRNAs) or antisense oligonucleotides to reduce protein levels and inhibit tumor growth in preclinical models (PubMed: 31435151). Targeting ZIC2 mRNA is particularly attractive in oncology because its expression is largely restricted in normal adult tissues, potentially offering a wide therapeutic window, although the delivery of nucleic acid-based drugs remains a significant challenge (PubMed: 30217957).
