Overview

Zinc finger SWIM-type containing 6 (ZSWIM6) encodes a highly conserved zinc finger protein with a SWIM (SWI2/SNF2 and MuDR) domain that is predicted to interact with DNA or proteins. It is critical for embryonic development, particularly the formation of craniofacial structures, the brain, and limbs. Variants in the gene are associated with disorders such as acromelic frontonasal dysostosis (characterized by frontonasal dysplasia, midline brain anomalies, and limb malformations) as well as neurodevelopmental syndromes. Functional studies show ZSWIM6 is expressed throughout the nervous system during early development, and its disruption is linked to altered Hedgehog pathway signaling and neuronal differentiation. Currently, there are no approved drugs targeting ZSWIM6, and it is not considered a classical therapeutic target, but rather a protein of developmental and genetic interest.
