Overview

Zinc fingers and homeoboxes protein 1 (ZHX1) is a nuclear transcriptional repressor encoded by the ZHX1 gene in humans. It is characterized by the presence of two C2H2-type zinc finger motifs and five homeobox DNA-binding domains, enabling it to bind DNA and repress gene transcription. ZHX1 forms homodimers and heterodimers (particularly with ZHX2 and ZHX3), and interacts with transcriptional regulators, including the A subunit of nuclear factor-Y (NF-YA). ZHX1 is predominantly localized in the nucleus and functions as a corepressor, participating in the regulation of cell differentiation—particularly in neural and hematopoietic lineages—and in the specification of cardiac progenitors. Dysregulation or aberrant expression of ZHX1 is associated with multiple diseases, including various cancers and kidney disorders. No therapeutic drugs are known to target ZHX1 directly as of the latest available literature.
