Overview

Zinc phosphodiesterase ELAC protein 1 (ELAC1) is a cytosolic and nucleoplasmic enzyme that functions as a tRNA 3'-processing endoribonuclease, specifically cleaving the 3' trailer sequence in tRNA precursors[1][4][5][7]. ELAC1 plays a critical role in tRNA maturation, distinct from the longer isoform ELAC2 which carries out similar processes in the nucleus and mitochondria[1]. ELAC1 is also implicated in the rescue and recycling of tRNA molecules during ribosome-associated quality control, removing aberrant 2',3'-cyclic phosphates and enabling re-addition of the terminal CCA sequence needed for functional tRNA[1][3][4]. ELAC1 belongs to the RNase Z family and contains zinc-binding motifs characteristic of phosphodiesterases[6][7]. There are no significant reports of drug interactions, biomarker status, or safety concerns, and it is not currently considered a major therapeutic target in human disease[4][5][6].
