Overview

Zinc transporter 5 (SLC30A5) is a membrane transporter responsible for mediating cellular zinc efflux and sequestration into organelles, primarily functioning in the early secretory pathway including the Golgi apparatus and vesicular compartments[1][2][3][4][5][6]. It forms heterodimers with zinc transporter 6 (SLC30A6), acting as a proton-coupled zinc antiporter critical for activation and folding of numerous zinc-dependent enzymes, such as alkaline phosphatases, and for the storage and secretion of insulin in pancreatic beta cells[1][3]. SLC30A5 is selectively essential for mast cell–mediated delayed-type allergic responses, influencing immune signaling by regulating the availability of zinc required for protein kinase C and NF-κB activation[4]. Alterations in its function have been associated with congenital disease, immune dysregulation, and possibly cancer, highlighting its significance for both basic biology and potential therapeutic targeting[3][4][5][6].
