Overview

ZNF496 divergent transcript (ZNF496-DT, XLOC_000647) is a long non-coding RNA transcribed from the genomic region adjacent to the ZNF496 protein-coding gene. It is not a protein, receptor, or enzyme, but rather a regulatory RNA. ZNF496-DT has been identified as downregulated in pancreatic cancer tissue and cell lines, and its increased expression suppresses cancer cell proliferation, invasion, and EMT by negatively regulating the nearby gene NLRP3. Clinically, lower expression of ZNF496-DT is associated with worse survival outcomes and higher lymph node metastasis rates in pancreatic cancer. ZNF496-DT is classified as a divergent transcript lncRNA, meaning it is transcribed from a region close to a protein-coding gene but in the opposite direction, serving a potential regulatory role. The gene is not considered a canonical therapeutic target such as a receptor, enzyme, or transporter. No known drugs target ZNF496-DT directly, nor have mechanisms of action or safety concerns been described for targeting it. Its role in cancer is as a suppressor, and not as a classical disease-driving target; thus, it is more of a biomarker or candidate lncRNA for research. The protein-coding gene ZNF496, located nearby, is a zinc finger protein involved in DNA-binding and transcriptional regulation, but it is distinct from the divergent transcript.
