ZNF503 antisense RNA 2 (ZNF503-AS2) is a long non-coding RNA (lncRNA) gene that is highly expressed in glioma, especially in high-grade and mesenchymal subtypes, and is strongly associated with poor prognosis in glioma patients[1]. Functionally, ZNF503-AS2 shapes the tumor immune microenvironment by promoting infiltration of immunosuppressive cells (macrophages, neutrophils, Tregs, MDSCs) and correlates positively with immune checkpoint molecules, stroma activation, and inflammatory response, potentially leading to reduced response to immunotherapy[1]. Knockdown of ZNF503-AS2 inhibits glioma cell proliferation, induces G2/M cell cycle arrest, stimulates apoptosis, and impairs cell migration and invasion[1]. ZNF503-AS2 is correlated with epigenetic regulatory pathways (methylation, mismatch repair, RNA modification) and may influence oncogenic transformation and immune evasion in tumor cells[1]. These roles suggest that ZNF503-AS2 is both a prognostic biomarker and a potential novel therapeutic target in cancer, especially in glioma[1][2][3].
