Overview

ZNF561 antisense RNA 1 is a long non-coding RNA located on human chromosome 19p13.2. It does not encode a protein, but regulates gene expression post-transcriptionally. In colorectal cancer, ZNF561-AS1 is overexpressed and acts as an oncogenic factor by sequestering miR-26a-5p and miR-128-3p, thereby promoting the upregulation of SRSF6, a serine-arginine-rich splicing factor. This stimulates cell proliferation, supports cell cycle progression, and suppresses apoptosis in cancer cells. ZNF561-AS1 may have tissue-specific roles, functioning as a tumor promoter in some contexts and a suppressor in others. No direct pharmacological agents currently target ZNF561-AS1, but its expression level is being explored as a biomarker and its inhibition represents a potential therapeutic strategy for cancers exhibiting dysregulation of this lncRNA

Mechanism of action

Inhibition of lncRNA-mediated miRNA sponging, suppressing downstream oncogenic pathways

03

Biological functions

Gene expression regulation (acts as a competitive endogenous RNA (ceRNA), sponging microRNAs)Cell proliferation (promotes cancer cell survival and growth in colorectal cancer)Cell cycle regulation (upregulates cell cycle promoting factors)Apoptosis (knockdown increases apoptosis in cancer cells)Other (post-transcriptional regulation of mRNA stability via miRNA interactions)
04

Disease associations

Cancer (most extensively studied in colorectal cancer and other malignancies)Other (possible role varies by tissue context—as an oncogene or tumor suppressor)
05

Biomarkers

Elevated expression of ZNF561-AS1 is a candidate biomarker for colorectal cancer prognosis and progression; possible association with SRSF6 and altered miR-26a-5p/miR-128-3p levels
