ZNF567 divergent transcript (ZNF567-DT), also known as LINC01534, is a long noncoding RNA (lncRNA) that does not encode protein but has regulatory roles in gene expression and cell physiology. It has been shown to maintain cancer cell stemness by suppressing the endoplasmic reticulum (ER) stress response, particularly through the PERK/eIF2α signaling pathway, in colorectal cancer. High levels of ZNF567-DT expression are correlated with increased tumor cell proliferation, invasion, cell cycle progression, and reduced apoptosis, and are associated with a poor prognosis in colorectal cancer patients. In addition to its role in cancer, ZNF567-DT (LINC01534) regulates inflammatory responses via the NF-κB pathway, acting as a competing endogenous RNA (ceRNA) that modulates the miR-135b-5p/PTPRT axis, and is implicated in pathological inflammation seen in conditions such as aseptic loosening after hip replacement surgery. Current data do not indicate this lncRNA is a direct therapeutic target for existing drugs, nor are there known drug interactions or mechanistic inhibitor classes targeting it[2][3][4].
