Overview

ZNF582 antisense RNA 1 (ZNF582-AS1, also called ZNF582 divergent transcript) is a long non-coding antisense RNA located on chromosome 19q13.43. It is transcribed from a locus adjacent to the sense gene ZNF582 in a head-to-head orientation, sharing a common promoter but with opposite transcriptional direction. ZNF582-AS1 is downregulated in several cancers, including breast and esophageal cancer, often through promoter hypermethylation. High levels of ZNF582-AS1 suppress tumor cell proliferation, colony formation, migration, and invasion, and promote apoptosis. These effects are partially mediated by regulation of oncogenic pathways (e.g., HER2/ERBB2 signaling) and may involve molecular sponging of oncogenic microRNAs (e.g., hsa-miR-940 in breast cancer). ZNF582-AS1 may act as an epigenetic biomarker for cancer diagnosis, prognosis, and therapeutic response, but is not itself a typical drug target. Restoration of its expression may be achieved via inhibition of promoter methylation. It primarily acts through regulation of gene expression, affecting its nearby protein-coding sense gene, ZNF582, and modulating key cellular functions involved in tumor biology.
