Overview

ZNF674 antisense RNA 1 (ZNF674-AS1) is a long non-coding RNA gene located on the X chromosome (chrX:46,545,438-46,564,575 in GRCh38) and does not encode a protein[1][5]. ZNF674-AS1 is primarily studied for its regulatory role in cancer biology. It acts through epigenetic and post-transcriptional mechanisms, including interaction with proteins such as EZH2 and ALDOA, to modulate the expression of neighboring or pathway genes (notably CHST7), alter glycolytic metabolism, and influence cellular processes such as proliferation, apoptosis, migration, invasion, and drug resistance[2][4][6]. Its expression is frequently reduced in various cancers, and lower levels are associated with poorer prognosis and increased chemoresistance, especially to oxaliplatin in gastric cancer and to cisplatin in other tumor types[2][3][4]. Overexpression or restoration of ZNF674-AS1 resensitizes cancer cells to chemotherapy, suggesting its potential role as a biomarker for prognosis and therapy response, but it is not a classical therapeutic target or receptor in itself[2][3][4].
