Zona pellucida glycoproteins are a family of four major glycoproteins (ZP1, ZP2, ZP3, ZP4) forming a thick glycoproteinaceous extracellular matrix around mammalian oocytes. This matrix is essential for species-specific sperm binding, triggering the acrosome reaction for fertilization, and preventing polyspermy. Its functions extend to embryonic protection, proper preimplantation development, and regulation of timing of blastocyst implantation. The ZP glycoproteins possess a conserved ZP domain that enables polymerization into a meshwork, with each member contributing distinct aspects to sperm binding and matrix structure (e.g., ZP3 as a primary sperm receptor, ZP1 as a structural cross-linker, ZP2 as a secondary sperm receptor, ZP4 as a facilitator of acrosome reaction). Dysfunction or disruption of these proteins leads to reproductive disorders or is exploited for non-hormonal contraceptive strategies.
For contraceptive vaccine development: Induction of immune response to inhibit ZP function, blocking sperm binding. For assisted fertilization: Targeting or supplementing ZP function to enhance or bypass sperm binding and fertilization.
