ZW10 interacting kinetochore protein (ZWINT) is a structural component of the outer kinetochore KNL1 complex, playing an essential adaptor role in chromosome segregation during mitosis and meiosis. ZWINT is required for proper assembly and function of the spindle assembly checkpoint and ensures faithful kinetochore-microtubule interactions. The protein acts by recruiting the RZZ complex and mitotic checkpoint proteins to the kinetochore and is regulated by post-translational modifications such as phosphorylation by Aurora B kinase and targeted degradation by APC/C–Cdc20. ZWINT overexpression is associated with several cancers, where it contributes to tumor proliferation, invasiveness, and poor clinical outcomes, making it a candidate biomarker and possible therapeutic target in oncology. No drugs are currently known to interact directly with ZWINT, and its direct modulation remains experimental.
