Overview

Zyg-11 family member A, cell cycle regulator (ZYG11A), is a protein-coding gene involved in cell cycle regulation, especially during meiosis, and in the regulation of cell proliferation and apoptosis[1][2][4][5]. ZYG11A is part of the ZYG11 gene family and is structurally related to ZYG11B, which functions as a substrate recruitment subunit for a cullin-2-based E3 ubiquitin ligase complex[1][2]. Dysregulation of this ubiquitin ligase system is linked to cancer and other diseases. ZYG11A itself has been implicated both as a potential oncogene (e.g., in non-small cell lung cancer) and as a candidate tumor suppressor (e.g., in ovarian cancer, where its low expression correlates with higher tumor grade)[2]. Its expression is modulated by the IGF1-p53 axis; IGF1 can increase or decrease ZYG11A expression depending on the status of p53[2]. Mutations affecting ZYG11A can cause cell cycle arrest, such as G2/M arrest in pancreatic β-cells, supporting a role in growth and metabolic regulation[2]. No drugs are currently known to target ZYG11A directly, and there are no established safety concerns related to therapeutic development at present. However, ZYG11A is emerging as a candidate biomarker in cancer, particularly epithelial ovarian cancer, where its levels correlate with disease grade and potentially prognosis[2].
