Overview

Zymogen granule protein 16 is a carbohydrate-binding lectin encoded by the ZG16 gene, predominantly expressed in secretory tissues such as the colon, endometrium, and spleen[2][5]. It serves as a linker between secretory proteins in the trans-Golgi network and the zymogen granule membrane, facilitating granule formation and proper protein trafficking[5][7]. ZG16 mediates defense against Gram-positive bacteria by supporting mucosal barrier integrity, and its downregulation is associated with colorectal cancer and other diseases affecting secretory and mucin-producing cells[2][3][5]. It features a jacalin-like lectin domain and a flexible C-terminal CXXC motif with redox activity, interacting with oxidases such as quiescin sulfhydryl oxidase 1 and protein disulfide isomerase[3]. Disruption of ZG16 impacts host defense, barrier function, and is implicated in several genetic and autoimmune diseases.
