αCD276 CAR-DNT cells are an experimental allogeneic cell therapy consisting of double-negative T (DNT) cells (CD3+CD4-CD8-) engineered with a second-generation chimeric antigen receptor (CAR) containing a 4-1BB costimulatory domain targeting CD276 (B7-H3). Developed by researchers at the University of Calgary, these cells are designed to treat glioblastoma (GBM) by addressing spatial heterogeneity. DNT cells are utilized for their innate anti-tumor cytotoxicity and their suitability for allogeneic use without inducing graft-versus-host disease (GvHD). In preclinical models, these cells have shown the ability to target both the tumor core and the infiltrative edge, potentially overcoming the limitations of conventional CAR-T therapies in heterogeneous brain tumors.
