12C2 is a humanized monoclonal antibody (hIgG1) that targets LIGHT (TNFSF14), a ligand in the tumor necrosis factor receptor (TNFR) family. Developed by the University of Bern in collaboration with Argenx, 12C2 is designed to block the interaction between LIGHT and the lymphotoxin beta receptor (LTbR). This signaling pathway is critical for the maintenance and expansion of leukemia stem cells (LSCs) in acute myeloid leukemia (AML). Research indicates that LIGHT is expressed on LSCs but not on normal hematopoietic stem cells, making it a selective target. By inhibiting LIGHT/LTbR signaling, 12C2 reduces LSC stemness and promotes differentiation. Furthermore, 12C2 can be modified using POTELLIGENT technology to enhance antibody-dependent cell-mediated cytotoxicity (ADCC), enabling NK cells to eliminate LIGHT-expressing leukemic cells.
