Overview

131I-Lym-1 is a radiopharmaceutical conjugate consisting of the murine monoclonal antibody Lym-1 labeled with the radioisotope Iodine-131. Lym-1 is an IgG2a antibody that specifically targets a unique epitope of the HLA-DR antigen, which is highly up-regulated on the surface of malignant B-lymphocytes in various hematologic cancers. The drug functions as a radioimmunotherapy (RAIT) agent; once the antibody binds to the target cells, the Iodine-131 delivers cytotoxic beta radiation (and gamma radiation for imaging) to the tumor site. Primarily investigated for the treatment of chemotherapy-resistant non-Hodgkin's lymphoma (NHL) and chronic lymphocytic leukemia (CLL), it demonstrated the ability to induce durable remissions. Clinical development identified dose-limiting thrombocytopenia and the common development of human anti-mouse antibodies (HAMA) as significant safety considerations.
