20-19 Tan BB06oz CAR T is an autologous, bispecific chimeric antigen receptor (CAR) T-cell therapy developed by Roswell Park Cancer Institute. It is designed to simultaneously target two B-cell antigens, CD19 and CD20, to reduce the risk of antigen escape, a common cause of relapse in single-target CAR-T therapies. The CAR construct is unique in its incorporation of tandem (Tan) signaling domains, specifically utilizing both a 4-1BB costimulatory domain and a mutated CD28 costimulatory domain to enhance T-cell persistence and potency. It is currently being evaluated in Phase I clinical trials for patients with relapsed or refractory CD19+ aggressive hematologic malignancies, including various types of B-cell lymphoma.
