Overview

20BBzT2 is a chimeric antigen receptor (CAR) T-cell therapy currently in preclinical development by Wellington Zhaotai for the treatment of B-cell acute lymphoblastic leukemia (B-ALL). The therapy involves engineering T cells to express a CAR that specifically targets the CD20 antigen, a protein widely expressed on the surface of mature B cells and most B-cell malignancies. The construct features a second-generation CAR design, incorporating a 4-1BB (CD137) co-stimulatory domain and a CD3 zeta signaling domain. The inclusion of the 4-1BB domain is intended to enhance the metabolic profile, expansion, and long-term persistence of the T cells following infusion, potentially providing a more durable anti-tumor response compared to first-generation CARs. 20BBzT2 aims to provide a targeted therapeutic option for patients with CD20-positive B-cell leukemias.
