4SCAR-T is a fourth-generation safety-designed chimeric antigen receptor (CAR) T-cell therapy developed by Shenzhen Geno-Immune Medical Institute. This innovative cell therapy involves genetically modifying autologous T-lymphocytes to express a CAR that targets specific tumor antigens. The 4SCAR design incorporates multiple co-stimulatory signaling domains, including CD28, CD137 (4-1BB), and CD27, to enhance T-cell activation, proliferation, and persistence. A key safety feature of 4SCAR-T is the inclusion of an inducible caspase 9 (iCasp9) suicide gene, which allows for the controlled elimination of CAR-T cells in vivo if severe adverse events, such as cytokine release syndrome (CRS), occur. This design aims to improve the safety profile while maintaining potent anti-tumor efficacy against various cancers by targeting specific tumor-associated antigens like GD2, FRa, CD19, CD20, CD22, CD30, CD70, CD123, and PSMA.
