Overview

4SCAR20 is a fourth-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD20 antigen, developed by the Shenzhen Geno-Immune Medical Institute for the treatment of B-cell malignancies. The therapy utilizes autologous T cells modified via lentiviral transduction to express a CAR construct featuring multiple costimulatory domains (CD28 and CD27) and the CD3ζ signaling chain, designed to enhance T-cell activation, expansion, and long-term persistence. A key feature of the 4SCAR platform is the inclusion of an inducible caspase 9 (iCasp9) safety switch, which allows for the rapid elimination of CAR-T cells in the event of severe adverse toxicities. 4SCAR20 is frequently investigated in clinical trials as part of a multi-target approach, often combined with 4SCAR19, to mitigate antigen escape and improve outcomes in patients with relapsed or refractory B-cell hematological cancers.
