9-ING-41 + ruxolitinib

9-ING-41 + ruxolitinib is an investigational combination therapy being evaluated for the treatment of advanced myelofibrosis. 9-ING-41 is a first-in-class, intravenously administered, maleimide-based, small molecule inhibitor of glycogen synthase kinase-3 beta (GSK-3β), with anti-cancer and anti-fibrotic activity[2][3]. Ruxolitinib is an oral Janus kinase (JAK) 1/2 inhibitor used to treat myelofibrosis; it is marketed as Jakafi[1]. Pre-clinical data suggest synergy between these agents in myelofibrosis, with 9-ING-41 potentially reducing ruxolitinib-induced myelosuppression and/or enabling optimal dosing of ruxolitinib[2][5].
