AAK1 (AP2-associated protein kinase 1) inhibitors are a class of compounds designed to block the activity of the AAK1 enzyme, a serine/threonine kinase that plays a critical role in clathrin-mediated endocytosis. AAK1 regulates the internalization of cell surface receptors by phosphorylating the mu2 subunit of the AP-2 adaptor complex. In the context of oncology, research suggests that AAK1 inhibition can enhance the trafficking and accumulation of T cells within solid tumors, potentially overcoming resistance to immunotherapies. However, preclinical studies have also indicated a complex antagonistic relationship where AAK1 inhibition may diminish the therapeutic efficacy of PD-1 blockade. Beyond oncology, AAK1 inhibitors are primarily being developed for the treatment of neuropathic pain, where they are thought to modulate the endocytosis of channels and receptors involved in pain signaling pathways.
