ABBV-438 is an investigational bispecific T-cell engager (TCE) developed by AbbVie for the treatment of relapsed or refractory multiple myeloma. The molecule is engineered to bind simultaneously to G protein-coupled receptor class C group 5 member D (GPRC5D), an antigen highly and selectively expressed on the surface of malignant plasma cells, and the CD3 subunit of the T-cell receptor complex. By bridging these two targets, ABBV-438 facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent release of cytotoxic granules that induce the targeted lysis of GPRC5D-positive myeloma cells. It is currently undergoing Phase 1 clinical evaluation as a monotherapy in patients who have received multiple prior lines of therapy.
