Overview

ABBV-599 is a novel fixed-dose oral combination of two small molecule inhibitors: elsubrutinib, a selective Bruton's tyrosine kinase (BTK) inhibitor, and upadacitinib, a Janus kinase 1 (JAK1) inhibitor. Developed by AbbVie, this investigational drug targets non-overlapping signaling pathways implicated in autoimmune diseases such as systemic lupus erythematosus (SLE) and rheumatoid arthritis. The rationale for combining these agents is to enhance efficacy by dual inhibition of BTK and JAK1 pathways while maintaining an acceptable safety profile. Clinical trials have demonstrated that ABBV-599 can significantly reduce disease activity and flares in SLE patients compared to placebo or monotherapy with either agent[2][3][5][6][7][8].
